In vitro generation of human monocyte-derived dendritic cells methodological aspects in a comprehensive review Anais da ABC (AABC)
CECHIM,GIOVANA; CHIES,JOSÉ A.B..
Abstract: Dendritic cells (DCs) initiate and shape both innate and adaptive immune responses. They are specialized in antigen presentation to naive T cells, thereby orchestrating the T cell immune responses. Human peripheral blood and tissues contain several subsets of phenotypically and functionally distinct DCs, which promote interactions between the external environment and lymphoid organs. Because of the difficulty in purifying these cells, in vitro studies only became more frequent when Frederica Sallusto and Antonio Lanzavecchia developed a method to generate DCs from blood monocytes in vitro. Nowadays a wide range of biotechnological innovations has allowed the study of DCs and their precursors in the most diverse situations faced by the immune...

In vitro proliferation and differentiation of hepatic oval cells and their potential capacity for intrahepatic transplantation BJMBR
Li,Z.; Chen,J.; Li,L.; Ran,J.H.; Liu,J.; Gao,T.X.; Guo,B.Y.; Li,X.H.; Liu,Z.H.; Liu,G.J.; Gao,Y.C.; Zhang,X.L..
Hepatic oval cells (HOCs) are recognized as facultative liver progenitor cells that play a role in liver regeneration after acute liver injury. Here, we investigated the in vitro proliferation and differentiation characteristics of HOCs in order to explore their potential capacity for intrahepatic transplantation. Clusters or scattered HOCs were detected in the portal area and interlobular bile duct in the liver of rats subjected to the modified 2-acetylaminofluorene and partial hepatectomy method. Isolated HOCs were positive for c-kit and CD90 staining (99.8% and 88.8%, respectively), and negative for CD34 staining (3.6%) as shown by immunostaining and flow cytometric analysis. In addition, HOCs could be differentiated into hepatocytes and bile duct...

Infectious bronchitis virus: detection and vaccine Strain differentiation by semi-nested RT-PCR Rev. Bras. Ciênc. Avic.
Okino,CH; Montassier,MFSM; Givisiez,PEN; Furuyama,CRAG; Brentano,L; Montassier,HJ.
A semi-nested reverse transcription-polymerase chain reaction (Semi-N-RT-PCR) was developed and used to detect the S glycoprotein gene of infectious bronchitis virus (IBV) strains and to discriminate H120 vaccine strain from other strains. Viral RNA was extracted from the allantoic fluid of chicken embryos and from tissues of chickens experimentally infected with different strains of IBV. Amplification and identification of the viral RNA was performed using two sets of primers complementary to a region of the S glycoprotein gene in the Semi-N-RT-PCR assay. The pair of primers used in the first PCR consisted of universal oligonucleotides flanking a more variable region of S1-S2 gene. The second primer pair was used in the Semi-N-RT-PCR and was comprised of...

Intramyocardial implantation of differentiated rat bone marrow mesenchymal stem cells enhanced by TGF-β1 improves cardiac function in heart failure rats BJMBR
Lv,Y.; Liu,B.; Wang,H.P.; Zhang,L..
The present study tested the hypotheses that i) transforming growth factor beta 1 (TGF-β1) enhances differentiation of rat bone marrow mesenchymal stem cells (MSCs) towards the cardiomyogenic phenotype and ii) intramyocardial implantation of the TGF-β1-treated MSCs improves cardiac function in heart failure rats. MSCs were treated with different concentrations of TGF-β1 for 72 h, and then morphological characteristics, surface antigens and mRNA expression of several transcription factors were assessed. Intramyocardial implantation of these TGF-β1-treated MSCs to infarcted heart was also investigated. MSCs were initially spindle-shaped with irregular processes. On day 28 after TGF-β1 treatment, MSCs showed fusiform shape, orientating parallel with one...

Market Structure and Competition in Food Retail: Some Evidences from Brazil AgEcon
Monterio, G.F.A.; Farina, Elizabeth Maria Mercier Querido; Nunes, R..
The paper analyzes competition among supermarkets in Brazil. In contrast to part of the economic literature which suggests that the fast growth of big supermarket chains would destroy independent, medium and small supermarkets, the paper argues that big supermarket chains can coexist with different formats of independent food retailing. As a result, competition in food retail is complex and cannot be described as a simple Darwinian process of market concentration. The analysis is divided in two parts. In the first part, the competition between hypermarkets and supermarkets is examined. Evidences for the district of Sao Paulo, Brazil, suggest that these retailers form separate markets. The second part is focused on neighborhood supermarkets. The results...

Molecular differentiation between Salmonella enterica subsp enterica serovar Pullorum and Salmonella enterica subsp enterica serovar Gallinarum BJM
Ribeiro,Simone Alves Mendes; Paiva,Jaqueline Boldrin de; Zotesso,Fábio; Lemos,Manoel Victor Franco; Berchieri Júnior,Ângelo.
S. Pullorum (SP) and S. Gallinarum (SG) are very similar. They are the agents of pullorum disease and fowl typhoid, respectively, and the two diseases are responsible for economic losses in poultry production. Although SP and SG are difficult to be differentiated in routine laboratory procedures, the ability to metabolize ornithine is a biochemical test that may be used to achieve this aim. While SP is able to decarboxylate this amino acid, SG is not. However, the isolation of strains showing atypical biochemical behavior has made this differentiation difficult. One of the genes associated with the metabolization of the amino acid ornithine is called speC, and is found in both serovars. The analysis of 21 SP and 15 SG strains by means of PCR did not enable...

Molecular differentiation of Salmonella Gallinarum and Salmonella Pullorum by RFLP of fliC gene from Brazilian isolates Rev. Bras. Ciênc. Avic.
Paiva,JB; Cavallini,JS; Silva,MD; Almeida,MA; Ângela,HL; Berchieri Junior,A.
Although Salmonella Pullorum and Salmonella Gallinarum cause different diseases in poultry, they are very similar. Both are non-motile and present the same somatic antigenic structure. They are differentiated by biochemical tests. Certain atypical strains are very difficult to distinguish. They do not produce the expected results when dulcitol and ornithine descarxboxylase tests are performed. Therefore, additional tests could be helpful. Many studies have chose the part I of the gene that encodes flagellin (fliC) to differentiate serotypes. Most Salmonella strains have two structural genes (fliC and fliB) that encode flagellins. Non-motile strains generally present these structural genes, but are not able to build a functional flagellum. It was...

Morphometrics and cytogenetics of Gracilinanus Agilis and Cryptonanus spp. (Didelphimorphia: Didelphidae) from central and northeastern Brazil Mastozool. neotrop.
Garcia,João P.; Oliveira,João A.; Corrêa,Margaret M. O.; Pessôa,Leila M..
Species of the didelphid genera Gracilinanus and Cryptonanus are morphologically and cytogenetically very similar. Several qualitative characters, some of which exhibit intraspecific polymorphisms, have been used to distinguish these genera, but more data are needed to characterize them better. Samples of G. agilis and Cryptonanus spp. from nine localities in central and northeastern Brazil were analyzed. Multivariate analyses of craniodental measurements and descriptive statistics of external body measurements indicate that G. agilis is conspicuously larger than Cryptonanus spp., and that general size is the main factor distinguishing these forms. Size differences can be combined with qualitative characters making the differentiation between G. agilis and...

Nkx-2.3 gene in mouse epidemial maturation Biol. Res.
MA,CHANG M.
In higher vertebrates, from amphibians to humans, epidemial maturation is a conserved developmental process. Using adult epidemial tissue and an established keratinocyte cell line, the mouse Nkx-2.3 homeobox gene was demonstrated, for the first time, to be expressed in mouse epidermal keratinocytes. Under the normal culture condition, the spontaneous aggregation phenomenon, a common initiation step of ES cell differentiation, and the induction of mouse adult K1 keratin, a marker of mature epidermal keratinocytes, were both observed in vitro when the Xenopus Nkx-2.3 gene was stably transfected into a mouse pluripotent P19 EC cell line. The induction of mouse K1 keratin by using its Xenopus orthologous gene in the mouse P19 cell implies that Nkx-2.3 may play...

Nuclear translocation of STAT3 by in vitro metreleptin administration causes lipolysis in human primary adipocytes BABT
Choi,Seung Kug; Park,Sunmi; Moon,Hyun-Seuk.
We utilized subcutaneous (SC)- and omental (OM)-derived human primary adipocytes (hPA) from obese male, and investigated whether synthetic analog of leptin, metreleptin, may regulate lipolysis via translocation of STAT3 to the nucleus. We observed that 50 ng/mL of metreleptin increases STAT3 phosphorylation in both SC- and OM-derived hPA. Importantly, we found for the first time that metreleptin is capable of trans-locating STAT3 to the nucleus and STAT3 blockade inhibits metreleptin-induced lipolysis. Our initial data provide novel insights into the role of STAT3 as probable mediator of the action of metreleptin in regulating metabolism.

Olivines in main-group pallasites: magma-ocean cumulates or partial melting residues? ArchiMer
Barrat, Jean-alix; Ferrière, L..
Main-group pallasites (MGPs) are meteorites mainly composed of Fe-Ni metal and olivines, the latter being considered as one of the largest sampling of extraterrestrial mantle material available for study on Earth. We analysed the rare earth element (REE) concentrations of olivines from six MGPs to understand better the processes of formation of their parent mantle. All the investigated samples display very low REE abundances, and enrichments in both light REEs and heavy REEs. We interpret the light REE enrichments as a fingerprint of terrestrial contamination. The least contaminated olivines have higher heavy REE enrichments than those inferred for olivines directly crystallised in a magma ocean. Such enrichments in heavy REEs are possible if the mantle of...

PD-1/PD-L1 regulates Treg differentiation in pregnancy-induced hypertension BJMBR
Jiang,Lai; Tang,Chaoliang; Gong,Yanping; Liu,Yujie; Rao,Jie; Chen,Suyu; Qu,Wanjun; Wu,Dabao; Lei,Lei; Chen,Ling.
Pregnancy-induced hypertension (PIH) causes significant maternal and fetal morbidity and mortality. A decreased number of regulatory T (Treg) cells is associated with the pathogenesis of PIH. The programmed cell death-1 (PD-1)/PD-ligand 1 (PD-L1) pathway is critical to normal pregnancy (NP) by promoting Treg cell development. However, the relationship between PD-1/PD-L1 and Treg differentiation in PIH has not been fully elucidated. In this study, venous blood was obtained from 20 NP and 58 PIH patients. Peripheral blood mononuclear cells (PBMCs) were isolated from venous blood. The levels of Treg-related cytokines (TGF-β, IL-10, and IL-35) in serum and PBMCs were measured by ELISA. The percentage of Treg cells in PBMCs was assessed by flow cytometry. The...

Retinoic acid and cAMP inhibit rat hepatocellular carcinoma cell proliferation and enhance cell differentiation BJMBR
Ionta,M.; Rosa,M.C.; Almeida,R.B.; Freitas,V.M.; Rezende-Teixeira,P.; Machado-Santelli,G.M..
Hepatocellular carcinoma (HCC) is the third highest cause of cancer death worldwide. In general, the disease is diagnosed at an advanced stage when potentially curative therapies are no longer feasible. For this reason, it is very important to develop new therapeutic approaches. Retinoic acid (RA) is a natural derivative of vitamin A that regulates important biological processes including cell proliferation and differentiation. In vitro studies have shown that RA is effective in inhibiting growth of HCC cells; however, responsiveness to treatment varies among different HCC cell lines. The objective of the present study was to determine if the combined use of RA (0.1 µM) and cAMP (1 mM), an important second messenger, improves the responsiveness of HCC...

Rhinovirus detection using different PCR-based strategies BJM
Silva,E.R.M.; Watanabe,A.S.A.; Carraro,E.; Perosa,A.H.S.; Granato,C.F.H.; Bellei,N.C.J..
Human rhinoviruses (HRVs) are the major cause of the common cold. HRVs were recently reclassified into the Enterovirus genus (HEV) in the Picornaviridae family. HRVs and other members of the HEV genus share many common features, including sense RNA genomes and partial nucleotide sequence identity. The aim of this study was to evaluate different HRV detection strategies. Samples from adults with acute respiratory infection (n = 291) who were treated in Sao Paulo Hospital (2001-2003) were tested using three assays. The first assay detected picornaviruses by RT-PCR and hybridization, the second detected rhinoviruses using RT-PCR/sequencing, and the third differentiated HRV from HEV using duplex semi-nested-RT-PCR. Analysis of the results obtained from the...

Roles of Hedgehog signaling pathway in adipogenic differentiation potential of porcine adipose-derived mesenchymal stem cells R. Bras. Zootec.
Fan,Caiyun; Zhang,Yunhai; Wang,Juhua; Cheng,Jianbo.
ABSTRACT This study examined the effects of purmorphamine and cyclopamine, classical agonists and inhibitors of the Hedgehog (Hh) signaling pathway, in the adipogenic differentiation of porcine adipose-derived mesenchymal stem cells (AMSC) to investigate the roles underlying adipogenic differentiation in AMSC. Porcine-derived AMSC were established, and the Hh signaling pathway was activated or inhibited by treatment with purmorphamine or cyclopamine. The adipogenic differentiation of the porcine AMSC was then analyzed by Oil Red O staining. The expression levels of Hh signaling pathway factors and adipogenic transcription factors were determined using quantitative real-time polymerase chain reaction and western blot analysis. We verified that the...

Sec61alpha synthesis is enhanced during translocation of nascent chains of collagen type IV in F9 teratocarcinoma cells after retinoic acid treatment BJMBR
Ferreira,L.R.; Velano,C.E.E.; Braga,E.C.; Paula,C.C.; Martéli Junior,H.; Sauk,J.J..
Nascent procollagen peptides and other secretory proteins are transported across the endoplasmic reticulum (ER) membrane through a protein-conducting channel called translocon. Sec61alpha, a multispanning membrane translocon protein, has been implicated as being essential for translocation of polypeptide chains into the cisterns of the ER. Sec61alpha forms a protein complex with collagen and Hsp47, an ER-resident heat shock protein that binds specifically to collagen. However, it is not known whether Sec61alpha is ubiquitously produced in collagen-producing F9 teratocarcinoma cells or under heat shock treatment. Furthermore, the production and utilization of Sec61alpha may depend on the stage of cell differentiation. Cultured F9 teratocarcinoma cells are...

Serotonin regulates osteoblast proliferation and function in vitro BJMBR
Dai,S.Q.; Yu,L.P.; Shi,X.; Wu,H.; Shao,P.; Yin,G.Y.; Wei,Y.Z..
The monoamine serotonin (5-hydroxytryptamine, 5-HT), a well-known neurotransmitter, also has important functions outside the central nervous system. The objective of this study was to investigate the role of 5-HT in the proliferation, differentiation, and function of osteoblasts in vitro. We treated rat primary calvarial osteoblasts with various concentrations of 5-HT (1 nM to 10 µM) and assessed the rate of osteoblast proliferation, expression levels of osteoblast-specific proteins and genes, and the ability to form mineralized nodules. Next, we detected which 5-HT receptor subtypes were expressed in rat osteoblasts at different stages of osteoblast differentiation. We found that 5-HT could inhibit osteoblast proliferation, differentiation, and...

Single embryoid body formation in a multi-well plate Electron. J. Biotechnol.
Ezekiel,Uthayashanker R; Muthuchamy,Mariappan; Ryerse,Jan S; Heuertz,Rita M.
Embryoid bodies (EB) formed from murine embryonic stem (ES) cells recapitulate many aspects of a developing embryo. Of specific importance, synchronous differentiation of EB recapitulates organ-specific development and is achieved in culture by formation of uniformly sized EB. The method described here demonstrates a simple and cost-effective way of generating EB from murine ES cells. Single EB are formed in a multi-well plate format and large numbers of EB are generated using a 96-well multi-well plate. Uniform single-sized EB formed in the multi-well are an ideal system for screening compounds and determining differentiation effects. Since EB contain all three germ layers, they are appropriate for studying small molecule effects on differentiation of ES...
